Clomipramine is a tricyclic antidepressant, the 3-chlorinated derivative of imipramine . Like imipramine, clomipramine potently inhibits serotonin and norepinephrine reuptake (Kis = 7.4 and 96 nM, respectively). It also is an antagonist at histamine, muscarinic acetylcholine, α1-adrenergic, and dopamine receptors (Kds = 31, 37, 38, and 190 nM for H1, M1, α1, and D2, respectively).Its main function is to block the reuptake of norepinephrine and 5-hydroxytryptamine in the central nervous system, and its blocking effect on the reuptake of 5-hydroxytryptamine is stronger. It also has antidepressant and anti-anxiety effects, as well as sedative and anti-cholinergic effects.

Application & Function

Clomipramine is used to treat obsessive compulsive disorder (OCD). It helps decrease thoughts that are unwanted or that don't go away (obsessions), and it helps reduce the urge to perform repeated tasks (compulsions such as hand-washing, counting, checking) that interfere with daily living.This medication belongs to a class of medications called tricyclic antidepressants. It works by restoring the balance of certain natural substances (serotonin, among others) in the brain.
