Rapamycin (RAPA) is a novel macrolide immunosuppressant, which is a white solid crystal with a melting point of 183-185 ºC. It is lipophilic and soluble in organic solvents such as ethanol, etc. It is extremely soluble in water and almost insoluble in ether. Initially used as a low toxicity antifungal drug, RAPA was discovered to have immunosuppressive effects in 1977. In 1989, it began to be tested as a new drug for treating organ transplant rejection. From animal experiments and clinical applications, it is a new type of immunosuppressant with good efficacy, low toxicity, and no nephrotoxicity. Nowadays, it is often used as a drug to maintain the immune function of transplanted organs (especially kidney transplantation) to slow down immune rejection after organ transplantation surgery. However, scientists have recently discovered another use for it: it can be used to treat Alzheimer's disease (senile dementia). What interests them is that the main component of rapamycin is also present in bacterial products isolated in the soil of Resurrection Island. The latest experiments have shown that the application of this substance to experimental mice can restore their ability to recognize defects.

Application&Function

In recent years, research has found that the target protein of rapamycin (mTOR), as an intracellular kinase, can induce various diseases due to abnormalities in its transmission pathway. Rapamycin, as a targeted inhibitor of mTOR, can treat tumors closely related to this pathway, including kidney cancer, lymphoma, lung cancer, liver cancer, breast cancer, neuroendocrine cancer and gastric cancer. Especially in the treatment of rare diseases such as LAM (lymphangioleiomyomatosis) and TSC (tuberous sclerosis), the effect is more significant, and LAM and TSC can also be considered as tumor diseases to some extent.
