Mesalazine, also known as 5-aminosalicylic acid, is an organic compound with the chemical formula C7H7NO3 and is the active ingredient in SASP for the treatment of ulcerative colitis. It has a significant inhibitory effect on intestinal wall inflammation. Mesalazine can inhibit the synthesis of inflammatory prostaglandins and the formation of inflammatory mediums leukotriene, which can significantly inhibit the inflammation of intestinal mucosa. Mesalazine inhibited prostaglandin synthesis and PGE2 release in human colon mucosa in a dose-dependent manner.
Application&Function

High levels of prostaglandin E2 (PGE2) and leukotriene B4 (LTB4) were found in the colon mucosa of patients with active UC. Mesalazine inhibited prostaglandin synthesis and PGE2 release in human colon mucosa in a dose-dependent manner. Mesalazine also inhibited the activity of fat oxidase in neutrophils. Two important inflammatory mediators, LTB4 and leukotriene, were dose-response inhibited by adding mesalazine to human colonic mucosal cells cultured. At high doses, mesalazine inhibits certain functions of human neutrophils, such as migration, threshing, phagocytosis, and oxygen free radical synthesis. In addition, mesalazine inhibits the synthesis of platelet motivity factors (PAFs), which play an important role in the development of inflammation. It works better on the connective tissue of the inflamed intestinal wall. It is used in ulcerative colitis, ulcerative proctitis and Crohn's disease.
Mesalazine can inhibit the synthesis of prostaglandins that cause inflammation and the formation of leukotriene, which can significantly inhibit the inflammation of intestinal mucosa.
