Trazodone is a triazole antidepressant that acts similarly to tricyclic and monoamine oxidase inhibitors (MAOIs). It has low toxicity to the cardiovascular system and no anticholinergic effect, making it more suitable for elderly and cardiovascular patients. The antidepressant mechanism selectively blocks the reabsorption of 5-HT and may have a weak blocking effect on NA reabsorption, but has no effect on dopamine, histamine, acetylcholine, and brain MAO activity. This product also has central sedative and mild muscle relaxant effects, but has no anti spasmodic or central excitatory effects. Has no effect on the heart, but can cause a decrease in blood pressure and is dose-dependent. Oral absorption is good, reaching its peak within 2 hours. After absorption, it is mostly distributed in the liver and kidneys, and through liver metabolism, the metabolites still have significant activity. Easy to pass through the blood-brain barrier, but rarely through the placental barrier. Metabolites are ultimately excreted through the kidneys. Rarely excreted in its original form. Renal dysfunction does not affect its excretion. T1/2 is 4.1 hours. Clinical trazodone can be used to treat depression. For patients with refractory depression who have not been treated with other antidepressants, this product is often effective. Trizodone hydrochloride belongs to the triazole class of antidepressants, mainly used to treat various types of depression, anxiety disorders with depressive symptoms, and emotional disorders in drug dependent individuals after withdrawal. Its exact mechanism of antidepressant action in the human body has not been fully elucidated, but it is generally believed that at therapeutic doses, its antidepressant pharmacological effect is to selectively inhibit the reabsorption of serotonin (5-HT) and may have a weak effect on preventing the reabsorption of norepinephrine, but it has no effect on dopamine, histamine, and acetylcholine, nor does it inhibit the activity of monoamine oxidase (MAO) in the brain. Trizodone hydrochloride has a strengthening effect on the behavioral changes induced by the precursor of 5-HT, 5-hydroxytryptamine acid.
