Gentamicin is an aminoglycoside drug that acts on ribosomes in bacteria, inhibits protein synthesis and destroys the integrity of bacterial membranes. The drug can be first passively diffused through the extracellular membrane pore protein and then transported through the cell membrane into the cell through the transport system, and irreversibly bound to the isolated ribosome 30S subunit, resulting in the destruction of the A-site, thus:
(1) preventing the correct localization of aminoacyl tRNA at a site, especially hindering the binding of methionyl tRNA, thus interfering with the assembly of functional ribosomes and inhibiting the formation of 70s initiating complex.
(2) the mismatching of tRNA and mRNA codon triad caused misreading of the genetic code by the 30S subunit of the intact ribosome, resulting in the wrong amino acid insertion into the protein structure, causes abnormal, nonfunctional protein synthesis.
(3) blocking the binding of the termination factor to a site, preventing the release of the synthesized peptide chain and preventing the dissociation of the 70s intact ribosome.
(4) it hinders the process of depolymerization and assembly of polyribosomes, resulting in the depletion of ribosomes in bacteria.
