Promethazine hydrochloride, also known as phenazine, is a commonly used phenothiazine antihistamine drug in clinical practice. Although adverse reactions have been reported in clinical practice, it is still widely used in primary hospitals due to its clear effects and low cost. Promethazine hydrochloride is the earliest discovered tricyclic antihistamine drug with a phenothiazine structure, which has cough stopping effects. It can competitively block the histamine H1 receptor to produce antihistamine effects, counteract histamine induced capillary dilation, and reduce its permeability. Therefore, it can alleviate coughing caused by stimulation of the trachea. Promethazine is a phenothiazine antihistamine drug that can also be used for antiemetic, anti motion, and sedative hypnotic purposes.
1. Antihistamine effect: Competes with histamine released by tissues for H1 receptors, can antagonize histamine's contraction or constriction of smooth muscles in the gastrointestinal tract, trachea, bronchi, or bronchioles, and relieve histamine's spasmodic and congested effects on bronchial smooth muscles.
2. Antiemetic effect: may be related to the inhibition of the emetic chemoreceptors in the medulla oblongata.
3. Anti motion sickness: It may act on the vestibular and vomiting centers as well as the midbrain sensory receptors through central anticholinergic properties, mainly blocking the excitation of cholinergic synaptic labyrinth impulses in the vestibular nucleus area.
4. Sedative and hypnotic effects: may be due to indirectly reducing the stress response of the brainstem reticular activating system

Application&Function

Promethazine hydrochloride can competitively block the Hl receptor in the limbs and produce antihistamine effects. It can counteract histamine induced capillary dilation, reduce its permeability, and alleviate wheezing caused by bronchial smooth muscle contraction. Its effect is stronger and more persistent than that of diphenhydramine hydrochloride. Suitable for various allergic diseases and vomiting during pregnancy.
1. Allergies to skin and mucous membranes: Suitable for long-term, seasonal allergic rhinitis, vasoconstrictive rhinitis, allergic conjunctivitis caused by exposure to allergens or food, urticaria, angioedema, allergic reactions to blood or plasma products, and skin scratch syndrome. If necessary, it can be used in combination as an adjuvant for this drug.
2. Motion sickness: prevention and treatment of motion sickness, seasickness, and airplane sickness.
3. Sedation and hypnosis: suitable for preoperative, postoperative, and obstetric use. In addition, it can also be used to alleviate fear in adults and children, resulting in a light sleep state.
