Valsartan is an antihypertensive drug of angiotensin II receptor antagonist, which can block the type I (AT1) receptor of angiotensin II, increase the plasma level of angiotensin II, stimulate the unsealed AT2 receptor, and counteract the AT1 receptor, so as to achieve the effect of expanding blood vessels and reducing blood pressure.

Application&Function

Valsartan is a non peptide, orally effective angiotensin(AT) receptor antagonist. It has high selectivity to type I receptor (AT1) and can antagonize competitively without any excitatory effect. It can also inhibit the release of aldosterol from adrenergic cells mediated by AT1 receptor, but valsartan has no inhibitory effect on the release caused by potassium, which also shows that valsartan has a selective effect on AT1 receptor. In vivo tests of various types of hypertensive animal models have shown that valsartan has a good antihypertensive effect, and has no significant impact on cardiac systolic function and heart rate. It has no hypotensive effect on animals with normal blood pressure. The bioavailability was 23%. The binding rate with plasma protein was 94%~97%. About 70% of them are excreted from the feces and 30% from the kidneys, both of which are in the original shape. t1/2 β About 9 hours. Taking it with food does not affect its efficacy. The blood pressure of patients with hypertension began to drop 2 hours after taking the drug once, and reached the maximum antihypertensive effect 4 to 6 hours later. The antihypertensive effect lasts for 24 hours. The blood pressure drop reached the maximum effect 2 to 4 weeks after continuous medication. It can be used together with hydrochlorothiazide to enhance the antihypertensive effect.
