Flucytosine has high antibacterial activity against Cryptococcus spp. and Candida spp. and has some antibacterial activity against staining fungi and a few Aspergillus spp. The antibacterial effect on other fungi are poor. It is an antibacterial agent and has bactericidal effect at high concentration. Because primary or secondary resistance to flucytosine is common, flucytosine is often used clinically in combination with other antifungal agents (often with amphotericin B).
The drug is a water-soluble nucleotide analogue and is well absorbed orally. It can enter the infected abdominal cavity, joint cavity and atrial fluid, and can cross the blood-brain barrier. The drug is widely distributed in liver, kidney, heart, spleen and lung tissues in concentrations greater than or equal to contemporaneous blood concentrations. The concentration of the drug in inflammatory CSF can reach 50%-100% of the blood concentration in the same period.
