Enfuvirtide, a pioneer in anti-HIV treatment for the last 7 years, is the first fusion inhibitor. Other anti-AIDS drugs work inside cells, preventing the virus from replicating inside cells. This drug works by preventing the virus from contacting and fusing with immune cells such as T cells, interfering with HIV-1 entering T cells, and preventing the immune system of AIDS patients from being destroyed by the virus, so it is also called fusion inhibitor
Uses
Enfuvirtide is a synthetic peptide HIV fusion inhibitor that binds to the viral envelope glycoprotein and prevents the conformational changes necessary for the virus to fuse with the cell membrane, thereby inhibiting HIV-1 replication. It is derived from a naturally occurring amino acid sequence in the HR2 domain of gp41. By simulating the activity of the HR2 domain and competitively binding the HR1 domain of gp41, it prevents the interaction between HR1 and HR2 and changes the configuration of gp41, thus preventing the fusion of virus and host cells.
